Hypophyseal prolactin levels in sexually mature female rats are 2–3 times greater than in sexually immature females. Sexual maturity has no effect on hypophyseal prolactin levels in the male rat.

Ovariectomy of sexually mature rats leads to a sharp decrease in hypophyseal prolactin. Replacement therapy with estradiol benzoate increases hypophyseal prolactin levels without, however, attaining the levels seen intact animals.

Estradiol benzoate leads to an increase in hypophyseal prolactin in sexually immature females, but does not affect the levels in sexually immature males.
